IL22 is an important cytokine involved in the intestinal defense mechanisms against microbiome. By using ileum-derived organoids, we show that the expression of anti-microbial peptides (AMPs) and anti-viral peptides (AVPs) can be induced by IL22. In addition, we identified a bacterial and a viral route, both leading to IL22 production by T cells, but via different pathways. Bacterial products, such as LPS, induce enterocyte-secreted SAA1, which triggers the secretion of IL6 in fibroblasts, and subsequently IL22 in T cells. This IL22 induction can then be enhanced by macrophage-derived TNFα in two ways: by enhancing the responsiveness of T cells to IL6 and by increasing the expression of IL6 by fibroblasts. Viral infections of intestinal cells induce IFNβ1 and subsequently IL7. IFNβ1 can induce the expression of IL6 in fibroblasts and the combined activity of IL6 and IL7 can then induce IL22 expression in T cells. We also show that IL22 reduces the expression of viral entry receptors (e.g. ACE2, TMPRSS2, DPP4, CD46 and TNFRSF14), increases the expression of anti-viral proteins (e.g. RSAD2, AOS, ISG20 and Mx1) and, consequently, reduces the viral infection of neighboring cells. Overall, our data indicates that IL22 contributes to the innate responses against both bacteria and viruses.
While the Municipality of Amsterdam wants to expand the electric vehicle public charging infrastructure to reach carbon-neutral objectives, the Distribution System Operator cannot allow new charging stations where low-voltage transformers are reaching their maximum capacity. To solve this situation, a smart charging project called Flexpower is being tested in some districts. Charging power is limited during peak times to avoid grid congestion and, therefore, enable the expansion of charging infrastructure while deferring grid investments. This work simulates the implementation of the Flexpower strategy with high penetration of electric vehicles, considering dynamic and local power limits, to assess the impact on both the satisfaction of electric vehicle users and the business model of the Charging Point Operator. A stochastic approach, based on Gaussian Mixture Models, has been used to model different profiles of electric vehicle users using data from the Amsterdam public electric vehicle charging infrastructure. Several key performance indicators have been defined to assess the impact of such charging limitations on the different stakeholders. The results show that, while Amsterdam’s existing public charging infrastructure can host just twice the current electric vehicle demand, the application of Flexpower will enable the growth in charging stations without requiring grid upgrades. Even with 7 times more charging sessions, Flexpower could provide a power peak reduction of 57% while supplying 98% of the total energy required by electric vehicle users.
Pre-eclampsia (PE) is a common and severe pregnancy complication and is associated with substantial perinatal morbidity and mortality in mothers and infants. The disease is often characterized by a non-specific presentation which makes it challenging for physician to diagnose PE during regular pregnancy check-ups. To date, there are no diagnostic tests on the market for detection of PE early in pregnancy (first trimester). In this project, we will develop a platform to sensitively analyse calcium-binding proteins (CBPs) which will unlock the full potential of CBPs as predictive PE markers. The technology will also be applicable for other diseases (e.g., dementia and cancer) where CBPs are also known to play a key role in disease pathophysiology. We will develop with phage display antibodies that can recognize calcium binding to specific motifs in proteins. To this end we will synthesize peptide motifs with and without calcium to select antibodies that are specific for calcium bound proteins. These antibodies will be validated for their clinical use. For this goal we will use serum samples from the Improved studie (EU subsidised study) to determine if we can recognize pre-eclampsia in a very early stage. This knowledge can lead to a better treatment of pregnant women suffering from this disease and also will probably increase the well-being for the baby born and the development further in life.

De sector voor vakantieparken en campings zal, net als alle andere sectoren, binnen dertig jaar nul-emissies van broeikasgassen moeten bereiken. De gemeente Goeree-Overflakkee en de Provincie Zuid-Holland zijn voornemens om een project op te zetten om de voor Goeree-Overflakkee belangrijke sector te helpen de energietransitie te maken. De gemeente en provincie willen graag antwoord op de vraag welke kennis en welke partijen nodig zijn om de duurzaamheidsdoelstelling zodanig in een beleidsproces te gieten dat de belangen van de vele stakeholders in deze sector samenkomen op een wijze dat de doelstelling gehaald wordt.This project entails an exploratory study that aims to map the technical route, scope and preconditions of the energy transition at holiday parks and campsites. The holiday park and camping sector, like all other sectors, will have to achieve zero greenhouse gas emissions within thirty years. The municipality of Goeree-Overflakkee and the Province of South Holland intend to set up a project to help this important sector on Goeree-Overflakkee to make the energy transition. The municipality and province would like an answer to the question of what knowledge and which parties are needed to put the sustainability objective into a policy process in such a way that the interests of the many stakeholders in this sector come together in a way that the objective is achieved.
Gender barriers are a complex problem, as they are created and maintained by multiple dimensions of our societal system; governments, the corporate world, and by society itself. Within the hospitality industry, one of the most people-oriented sectors there is, gender barriers are especially a problem. Although there is equality amongst the entire Dutch hospitality sector in general (48.2% women, (CBS, 2022)), only 17% of top-management positions within the 5 largest hotel-chains in the country are occupied by women (Hampshire Hotel Group, 2022; Accor Group, 2022; van der Valk International, 2022; NH Hotels, 2022; NIBC, 2022). With the hospitality industry revolving around people and experiences, it is of utmost importance that it represents the actual world-population and society. In order to address the current challenges the industry is facing, it is time to face the elephant in the room; why don’t women get included in top and senior management within the hospitality industry as much as men do? This trajectory aims to identify where gender barriers occur within the Dutch hospitality industry and accordingly develop, and test interventions (enablers) together with two of the largest hotel-chains in the Netherlands, in order to improve women career advancement. The first phases of the trajectory will focus on the entire Dutch hotel sector, while the intervention phase will only be executed in collaboration with NH Hotels and IHG. The final phase of the trajectory will explore the implications of the findings from the industry to hospitality management education. By enabling more women to advance their hospitality careers, this will have a large impact on the industry’s sustainability and resilience, and again positively impact wider society.
