Knowledge of the time of deposition is pivotal in forensic investigations. Recent studies show that changes in intrinsic fluorescence over time can be used to estimate the age of body fluids. These changes have been attributed to oxidative modifications caused by protein–lipid interactions. This pilot study aims to explore the impact of these modifications on body fluid fluorescence, enhancing the protein–lipid model system for age estimation. Lipid and protein oxidation markers, including protein carbonyls, dityrosine, advanced glycation end-products (AGEs), malondialdehyde (MDA), and 4-hydroxynonenal (HNE), were studied in aging semen, urine, and saliva over 21 days. Surface plasmon resonance imaging (SPRi), enzyme-linked immunosorbent assay (ELISA), and fluorescence spectroscopy were applied. Successful detection of AGE, dityrosine, MDA, and HNE occurred in semen and saliva via SPRi, while only dityrosine was detected in urine. Protein carbonyls were measured in all body fluids, but only in saliva was a significant increase observed over time. Additionally, protein fluorescence loss and fluorescent oxidation product formation were assessed, showing significant decreases in semen and saliva, but not in urine. Although optimization is needed for accurate quantification, this study reveals detectable markers for protein and lipid oxidation in aging body fluids, warranting further investigation.

The age estimation of biological traces is one of the holy grails in forensic investigations. We developed a method for the age estimation of semen stains using fluorescence spectroscopy in conjunction with a stoichiometric ageing model. The model describes the degradation and generation rate of proteins and fluorescent oxidation products (FOX) over time. The previously used fluorimeter is a large benchtop device and requires system optimization for forensic applications. In situ applications have the advantage that measurements can be performed directly at the crime scene, without additional sampling or storage steps. Therefore, a portable fiber-based fluorimeter was developed, consisting of two optimized light-emitting diodes (LEDs) and two spectrometers to allow the fluorescence protein and FOX measurements. The handheld fiber can be used without touching the traces, avoiding the destruction or contamination of the trace. In this study, we have measured the ageing kinetics of semen stains over time using both our portable fluorimeter and a laboratory benchtop fluorimeter and compared their accuracies for the age estimation of semen stains. Successful age estimation was possible up to 11 days, with a mean absolute error of 1.0 days and 0.9 days for the portable and the benchtop fluorimeters, respectively. These results demonstrate the potential of using the portable fluorimeter for in situ applications.

Semen traces are considered important pieces of evidence in forensic investigations, especially those involving sexsual offenses. Recently, our research group developed a fluorescence-based technique to accurately determine the age of semen traces. However, the specific compounds resonsible for the fluoresescent behaviour of ageing semens remain unknown. As such, in this exploratory study, the aim is to identify the components associated with the fluorescent behavior of ageing semen traces. In this investigation semen stains and various biofluorophores commonly found in body fluids were left to aged for 0, 2, 4, 7, 14 and 21 days. Subsequently, thin-layer chromatography (TLC) and ultra-performance liquid chromatography (UPLC) mass spectrometry were performed to identify the biofluorophores present in semen. Parasitoids lay their eggs in or on a host, usually another insect. During foraging, parasitoids can encounter insects that differ in terms of host suitability and quality. At one extreme end of this spectrum are non-hosts that are unsuitable for offspring development.Non-hosts are generally ignored but parasitization does occur and occasionally also results in egg deposition. Here, the authors investigate how oviposition in a non-host influences subsequent foraging behaviour of a parasitoid and whether this is mediated by learning. The study system consists of the endoparasitoid Cotesia glomerata and the presumed non-host caterpillar Mamestra brassicae.In the presence of Pieris brassicae hosts and/or their traces (frass), C. glomerata inserted its ovipositor into M. brassicae caterpillars. Eggs were deposited, but all eggs disappeared within 96 h, confirming the non-host status of M. brassicae. In contrast to the expectation, there was no memory retention after oviposition in a non-host and parasitoids did not alter their behaviour with respect to non-host contacts and ovipositions. Instead, C. glomerata became more motivated to forage on a non-host infested leaf.The authors propose that egg deposition in non-hosts by C. glomerata might be due to their high egg load, which is thought to make parasitoids less selective on host quality, especially when they have few reproductive opportunities. In such cases, fitness costs to individual females are low. Egg deposition in non-hosts might ultimately lead to host range expansion if parasitoids overcome the defence response of non-hosts over evolutionary time.

In this study, we experimentally investigated the material flows of coloured dyes in coalescing small inkjet printed droplets of different volume ratios. With two differently coloured dyes, one in each droplet, we were able to distinguish the time-resolved contributions in dye transport across the coalescence bridge due to convection and diffusion. Droplets with differently coloured dyes were inkjet printed onto a glass substrate at a sufficiently large pitch such that they do not touch each other. Under UV exposure, the wetting of the substrate improves, causing the droplets to coalesce. Filmed at 50 fps, the coalescence and mixing of the droplets of volume ratios 1:1, 2:1 and 4:1 was followed. For equally sized drops, the mixing of the dyes shows good agreement with a 1D approximation of Fick’s second law along the central axes of the coalescing droplets with a diffusion coefficient D = 2 9 10-9 m2 s-1. For unequally sized droplets, convective flows from the small to the large droplet were demonstrated. The convective flows increase in size with increasing volume ratio, but only enter the droplet over a small distance. Complete mixing of the dyes in the unequally sized droplets is only reached after a long period and is diffusion controlled. At the initial moment of coalescence of unequally sized droplets, a small convective flow is observed from the large into the small droplets. Further investigation in this phenomenon is recommended.

Fouling plays a major role in the Dairy industry. Five criteria: defined flow, no circulation, real factory product, defined product temperature and defined wall temperature, are used to review articles on this topic published between 2003 and 2020. To show the effect of those criteria in experiments, a simulation model is developed. For a good experimental design to measure fouling, the use of a dairy product in a tubular heater with a known developed flow is advised. The temperature-time history of the product and the wall temperature of the heater should be recorded. Circulation of a product will increase the fouling and decrease the flow. Although none of the reviewed articles complied to all criteria, 71% of the reviewed articles met at least two criteria. If not all criteria are met, the results are of less use for the application for process lines on industrial scale. A simulated computer model can be helpful.

During times of high activity by predators and competitors, herbivores may be forced to forage in patches of low‐quality food. However, the relative importance in determining where and what herbivores forage still remains unclear, especially for small‐ and intermediate‐sized herbivores. Our objective was to test the relative importance of predator and competitor activity, and forage quality and quantity on the proportion of time spent in a vegetation type and the proportion of time spent foraging by the intermediate‐sized herbivore European hare (Lepus europaeus). We studied red fox (Vulpes vulpes) as a predator species and European rabbit (Oryctolagus cuniculus) as a competitor. We investigated the time spent at a location and foraging time of hare using GPS with accelerometers. Forage quality and quantity were analyzed based on hand‐plucked samples of a selection of the locally most important plant species in the diet of hare. Predator activity and competitor activity were investigated using a network of camera traps. Hares spent a higher proportion of time in vegetation types that contained a higher percentage of fibers (i.e., NDF). Besides, hares spent a higher proportion of time in vegetation types that contained relatively low food quantity and quality of forage (i.e., high percentage of fibers) during days that foxes (Vulpes vulpes) were more active. Also during days that rabbits (Oryctolagus cuniculus) were more active, hares spent a higher proportion of time foraging in vegetation types that contained a relatively low quality of forage. Although predation risk affected space use and foraging behavior, and competition affected foraging behavior, our study shows that food quality and quantity more strongly affected space use and foraging behavior than predation risk or competition. It seems that we need to reconsider the relative importance of the landscape of food in a world of fear and competition.
