To accelerate differentiation between Staphylococcus aureus and Coagulase Negative Staphylococci (CNS), this study aimed to compare six different DNA extraction methods from 2 commonly used blood culture materials, i.e. BACTEC and Bact/ALERT. Furthermore, we analyzed the effect of reduced blood culture times for detection of Staphylococci directly from blood culture material. A real-time PCR duplex assay was used to compare 6 different DNA isolation protocols on two different blood culture systems. Negative blood culture material was spiked with MRSA. Bacterial DNA was isolated with: automated extractor EasyMAG (3 protocols), automated extractor MagNA Pure LC (LC Microbiology Kit MGrade), a manual kit MolYsis Plus, and a combination between MolYsis Plus and the EasyMAG. The most optimal isolation method was used to evaluate reduced bacterial culture times. Bacterial DNA isolation with the MolYsis Plus kit in combination with the specific B protocol on the EasyMAG resulted in the most sensitive detection of S.aureus, with a detection limit of 10 CFU/ml, in Bact/ALERT material, whereas using BACTEC resulted in a detection limit of 100 CFU/ml. An initial S.aureus load of 1 CFU/ml blood can be detected after 5 hours of culture in Bact/ALERT3D by combining the sensitive isolation method and the tuf LightCycler assay.

Matrix-assisted laser desorption/ionisation time of-flight mass spectrometry (MALDI-TOF MS) is a fast and reliable method for the identification of bacteria from agar media. Direct identification from positive blood cultures should decrease the time to obtaining the result. In this study, three different processing methods for the rapid direct identification of bacteria from positive blood culture bottles were compared. In total, 101 positive aerobe BacT/ALERT bottles were included in this study. Aliquots from all bottles were used for three bacterial processing methods, i.e. the commercially available Bruker's MALDI Sepsityper kit, the commercially available Molzym's MolYsis Basic5 kit and a centrifugation/washing method. In addition, the best method was used to evaluate the possibility of MALDI application after a reduced incubation time of 7 h of Staphylococcus aureus- and Escherichia coli-spiked (1,000, 100 and 10 colony-forming units [CFU]) aerobe BacT/ALERT blood cultures. Sixty-six (65%), 51 (50.5%) and 79 (78%) bottles were identified correctly at the species level when the centrifugation/washing method, MolYsis Basic 5 and Sepsityper were used, respectively. Incorrect identification was obtained in 35 (35%), 50 (49.5%) and 22 (22%) bottles, respectively. Gram-positive cocci were correctly identified in 33/52 (64%) of the cases. However, Gram-negative rods showed a correct identification in 45/47 (96%) of all bottles when the Sepsityper kit was used. Seven hours of pre-incubation of S. aureus- and E. coli-spiked aerobe BacT/ALERT blood cultures never resulted in reliable identification with MALDI-TOF MS. Sepsityper is superior for the direct identification of microorganisms from aerobe BacT/ALERT bottles. Gram-negative pathogens show better results compared to Gram-positive bacteria. Reduced incubation followed by MALDI-TOF MS did not result in faster reliable identification.

This article examines the ethics of tourism and cross-cultural communication between Western tourists and the local community in a Muslim country. Communicating with people who have different value systems and communication styles can contribute to various "culture shock situations" and to an increase in stereotypes and "stigma." The main goal of this research is to analyze the dress behavior of cruise tourists, applying the concept of "mindfulness" and secondly to analyze the voices and values of the resident community and of other tourists. Any apparent contradictions will be identified between local values, pretravel information, media social constructions, and tourist dress behaviors, and suggestions will be proposed about how to avoid culture shock situations. Two questionnaire surveys were conducted with German-speaking cruise tourists visiting two different destinations in the Sultanate of Oman during 2012 (N = 830) and 2013 (N = 235). In addition, in-depth interviews were conducted with members of the local community and cultural brokers as well as with tourists, onboard tour guides, and an onboard pastor. Moreover, government officials and the Assistant Grand Mufti were interviewed and pretravel information was studied. The results indicate that a mindless dress behavior has been facilitated by the type of information that is provided prior to travel and by cultural brokers, both on shore and on board, who do not make explicit reference to local dress codes. This approach promotes a concept of tolerance towards the tourist and an "accommodationist" and "laissez faire" attitude. On the other hand, tourist dress behaviors can be seen as a reflection of the posttourist, who is seeking individual authenticity and freedom. For the local community the increase in the number of mindless cruise tourists exceeds the level of acceptable tolerance in both places and has created "culture shock" situations. This research fills a major gap in applied research on the cruise tourist behaviors in a Muslim country and on crosscultural communication.

From Pubmed: " BACKGROUND: Antigen-specific immunotherapy (AIT) is a promising therapeutic approach for both cow's milk allergy (CMA) and peanut allergy (PNA), but needs optimization in terms of efficacy and safety. AIM: Compare oral immunotherapy (OIT) and subcutaneous immunotherapy (SCIT) in murine models for CMA and PNA and determine the dose of allergen needed to effectively modify parameters of allergy. METHODS: Female C3H/HeOuJ mice were sensitized intragastrically (i.g.) to whey or peanut extract with cholera toxin. Mice were treated orally (5 times/week) or subcutaneously (3 times/week) for three consecutive weeks. Hereafter, the acute allergic skin response, anaphylactic shock symptoms and body temperature were measured upon intradermal (i.d.) and intraperitoneal (i.p.) challenge, and mast cell degranulation was measured upon i.g. challenge. Allergen-specific IgE, IgG1 and IgG2a were measured in serum at different time points. Single cell suspensions derived from lymph organs were stimulated with allergen to induce cytokine production and T cell phenotypes were assessed using flow cytometry. RESULTS: Both OIT and SCIT decreased clinically related signs upon challenge in the CMA and PNA model. Interestingly, a rise in allergen-specific IgE was observed during immunotherapy, hereafter, treated mice were protected against the increase in IgE caused by allergen challenge. Allergen-specific IgG1 and IgG2a increased due to both types of AIT. In the CMA model, SCIT and OIT reduced the percentage of activated Th2 cells and increased the percentage of activated Th1 cells in the spleen. OIT increased the percentage of regulatory T cells (Tregs) and activated Th2 cells in the MLN. Th2 cytokines IL-5, IL-13 and IL-10 were reduced after OIT, but not after SCIT. In the PNA model, no differences were observed in percentages of T cell subsets. SCIT induced Th2 cytokines IL-5 and IL-10, whereas OIT had no effect. CONCLUSION: We have shown clinical protection against allergic manifestations after OIT and SCIT in a CMA and PNA model. Although similar allergen-specific antibody patterns were observed, differences in T cell and cytokine responses were shown. Whether these findings are related to a different mechanism of AIT in CMA and PNA needs to be elucidated."

Aim. Although cultural dimensions theory is a topical strand of quantitative cultural research, few intercultural simulation games use it. We present the design and review of the application of OASISTAN, an intercultural role-playing simulation game that is specifically based on cultural dimensions theory. Method. OASISTAN was first designed in 1999 for use in Master’s courses on cross-cultural management at Delft University of Technology in the Netherlands, attracting 20-23 year old students with a Bachelor degree in engineering and from various cultural backgrounds. Since its first design the game has been played approximately 45 times at Delft University of Technology in the Netherlands and three times at Harbin Institute of Technology in China in the years 2006-2008. We reviewed their experiences designing and facilitating OASISTAN since 1999. Results. The game has a no-tech role-play design and revolves around the geopolitically complex region of the Caspian Sea, specifically the fictional country of ‘Oasistan’. The game consists of students forming small teams of Oasistani, Western and non-Western public/private actors collaborating with each other to try and reach the common goal of oil exploration and production in this country. In total 15-30 students were involved. We found that OASISTAN allowed its players not only to intensely experience the difficulty and awkwardness of being confronted with cultural differences, but also to interpret and understand these differences through cultural dimensions. Students who played OASISTAN identified ten out of the 12 dimensions by Maleki and De Jong. The two dimensions that students were not able to identify are uncertainty avoidance and collaborativeness. Conclusion. OASISTAN shows how a game design field (i.e., intercultural simulation gaming) can be reinvigorated in light of new or updated scientific theories pertaining to the field’s subject matter (i.e., cultural dimensions). Several opportunities for future research are identified.

At present, leading international agencies, such as the United Nations Environmental Programme, are largely focused on what they claim to be ‘win-win’ scenarios of ‘sustainable development’ rhetoric. These combine social, economic and environmental objectives. However, as noted by the ‘Scientists’ Warning to Humanity’, environmental integrity is the essential precondition for the healthy functioning of social and economic systems, and thus environmental protection needs to be prioritized in policy and practice. Ecological sustainability cannot be reached without realizing that population growth and economic growth, with attendant increased rates of depletion of natural resources, pollution, and general environmental degradation, are the root causes of unsustainability. This article argues that to strategically address ecological unsustainability, the social, economic and political barriers to addressing the current economic model and population growth need to be overcome. Strategic solutions proposed to the current neoliberal economy are generic – namely, degrowth, a steady-state economy, and a ‘circular economy’. Solutions to demographic issues must be sensitive to the countries' cultural, social, political and economic factors to be effective as fertility differs from country to country, and culture to culture. As discussed here, Mediterranean countries have the lowest fertility in the world, while many countries in Africa, and some in Asia, South America have stable but consistently high birthrates. Dutch National Sports Organizations (NSFs) is currently experiencing financial pressures. Two indications for this are described in this paper i.e. increased competition in the sports sector and changes in subsidy division. Decreasing incomes from subsidies can be compensated with either increasing incomes from a commercial domain or increasing incomes from member contributions. This latter solution is gaining interest as a solution for the uncertainties. Many NSFs have therefore participated in a special marketing program in order to enlarge their marketing awareness and create a marketing strategy, in order to (re)win market share on the sports participation market and gain a more stable financial situation. This paper introduces my research related to the introduction of marketing techniques within NSFs and the change-over to become market oriented. An overview of existing literature about creating marketing strategies, their implementation, and market orientation is given. This outline makes obvious that the existing literature is not sufficient for studying the implementation of marketing techniques and market orientation within NSFs. Therefore, it shows the scientific relevance of my research. The paper concludes with the chosen research methodology.

This study empirically examines individual and organizational factors that influence expatriates’ cross-cultural adjustment and job performance. The study was a quantitative research from 117 Thai expatriates who work in Thai multinational companies (MNC) located in Indonesia. The results of the study indicated that financial perceived organizational support influence positively towards Thai expatriates’ overall cross-cultural adjustment in Indonesia. This study found that cross-cultural training influenced positively towards Thai expatriates’ adjustment. A causal relationship between the predicting variables of crosscultural adjustment and Thai expatriates’ job performance was not found. Results suggest important consequences for management strategies providing support to Thai expatriate employees increasing their adjustment in Indonesia. Keywords: Cross-Cultural Adjustment; Job

Hogeronderwijsinstellingen hebben verschillende redenen voor internationalisering. Een van de belangrijkste redenen vormt de ontwikkeling van internationale competenties van studenten. Van de instrumenten die zij daartoe inzetten wordt een verblijf in het buitenland om redenen van studie of stage als een van de sterkste beschouwd. De toegevoegde waarde van een studie of stage in het buitenland is onderzocht onder de groep van studenten van De Haagse Hogeschool die in de jaren 2009-2010 en 2011-2012 naar het buitenland is geweest. Daarbij is gebruik gemaakt van een online vragenlijst, interviews na terugkeer uit het buitenland en een 360 gradenfeedback. De uitkomsten wijzen over het algemeen op een toename van de persoonlijke,sociale en interculturele competenties van studenten als gevolg van hun verblijf in het buitenland, evenals hun beheersing van een vreemde taal. In mindere mate geldt dit voor hun academische en professionele competenties. Een buitenlandverblijf om reden van stage, in het bijzonder wanneer studenten daarbij ondergedompeld worden in de lokale cultuur buiten Europa, laat een sterkere leercurve zien dan een buitenlandverblijf om redenen van studie. Studenten leren meer wanneer zij goed worden voorbereid en begeleid. Studenten tonen zich echter kritisch met betrekking tot de informatievoorziening, voorbereiding en begeleiding vanuit de opleiding. In die zin is er ruimte voor verbetering met betrekking tot het faciliteren en begeleiden van studenten voorafgaand aan, tijdens en na terugkomst van hun verblijf in het buitenland. Niet alleen kan daarmee de ontwikkeling van internationale competenties versterkt worden, ook zou daarmee het aantal studenten dat kiest voor een verblijf in het buitenland, vergroot kunnen worden. ABSTRACT Institutes of higher education are internationalizing for various reasons. One of the most important reasons is to provide their students international competencies. Of the instruments they deploy, the most powerful one is considered to be a period abroad for study or internship. The surplus value of to study or an internship abroad was investigated in the group of students of The Hague University of Applied Sciences who went abroad in the academic years 2009-2010 and 2011-2012. This was done by means of an online questionnaire, post-return interviews and 360-degrees feedback. In both the questionnaire and the interviews the students invariably indicate a slight to strong increase in their personal, social, intercultural and linguistic competencies as the result of their study or internship abroad. They are slightly less positive about their academic and professional development. Internships, particularly when students have been submerged in the local culture outside Europe, produce a steeper learning curve than study in Europe. Students learn more when they are better prepared and guided. According to this study there is considerable room for improvement with regard to information about study and internships abroad, facilitation of students, preparation before and guidance during the stay abroad and clarity about the competencies to be acquired. These improvements could not only increase the number of students going abroad, but will enhance the acquisition of international competencies in those who go.
