In this article three different responses are taken as the starting point how different types of disruption could be dealt with. These responses—repair, bounce back and grow stronger—are combined with three disruptions (sea level rise, storm surge and heavy rainfall), and then tested in three case studies. The result of the investigation is that anti-fragility (grow stronger) is a preferential approach to create delta landscapes that become stronger under influence of a disruption. Anti-fragility is for this research subdivided in three main characteristics, abundance of networks, adaptivity and counterintuitivity, which are used to analyse the three case study propositions. The type of response, type of disruption, characteristic of anti-fragility and the qualities of the case study area itself determine the design proposition and the outcome. In all cases this approach has led to a stronger and safer landscape. The concept of anti-fragility impacts on the period before a disruption, during and also after the disruptive impact. This gives it a better point of departure in dealing with uncertain or unprecedented hazards and disruptions.

Dit proefschrift onderzoekt de manieren waarop affectieve reacties op lichamen met een handicap worden gerepresenteerd en hoe dit ons uitnodigt om deze lichamen esthetisch te lezen. Ik stel dat deze affectieve impact begrepen kan worden als een ‘affordance’, een term die ik gebruik om te beschrijven hoe de verschijning van en interactie met gehandicapte lichamen affectieve reacties teweegbrengt, zoals angst, verwondering of walging. Ik bestudeer de relatie tussen representatie en affectieve reacties via literatuur en andere kunstvormen. Door middel van close readings van literaire teksten en kunstwerken biedt dit proefschrift een alternatief voor het zogenaamde modeldenken - een benadering die categorisering benadrukt. In plaats daarvan stel ik een lezing voor die zich richt op hoe lichamelijke capaciteiten cultureel en sociaal vertaald worden in (on)vermogens. In tegenstelling tot taxonomische benaderingen die categoriseren en generaliseren, maakt deze methode het mogelijk om van het bijzondere naar het private te gaan. Kunstwerken, hoewel vatbaar voor generalisatie, benadrukken hun uniciteit en weerstaan categorisering. Door te analyseren hoe verschillende kunstvormen gehandicapte lichamen representeren, geeft dit proefschrift een nieuwe dimensie aan het begrijpen van onze emotionele reacties en de esthetische waardering van lichamelijke diversiteit.

Some patients in psychiatric treatment are considered extremely difficult to treat because of the disruptive nature of their relationships with treatment staff. In this paper, we describe and evaluate a specialist inpatient treatment program for these patients. Data were collected from medical records and daily reports of patients (n = 108). Pretest-posttest measurements were used to evaluate the treatment. The main treatment method consists of the provision of safety, structure, and cooperation. Treatment results show statistically significant changes from admittance to discharge. The collaborative and consistent manner in which nurses approach the patients is crucial for quality of care.

AimsGenetic hypertrophic cardiomyopathy (HCM) is caused by mutations in sarcomere protein-encoding genes (i.e. genotype-positive HCM). In an increasing number of patients, HCM occurs in the absence of a mutation (i.e. genotype-negative HCM). Mitochondrial dysfunction is thought to be a key driver of pathological remodelling in HCM. Reports of mitochondrial respiratory function and specific disease-modifying treatment options in patients with HCM are scarce.Methods and resultsRespirometry was performed on septal myectomy tissue from patients with HCM (n = 59) to evaluate oxidative phosphorylation and fatty acid oxidation. Mitochondrial dysfunction was most notably reflected by impaired NADH-linked respiration. In genotype-negative patients, but not genotype-positive patients, NADH-linked respiration was markedly depressed in patients with an indexed septal thickness ≥10 compared with <10. Mitochondrial dysfunction was not explained by reduced abundance or fragmentation of mitochondria, as evaluated by transmission electron microscopy. Rather, improper organization of mitochondria relative to myofibrils (expressed as a percentage of disorganized mitochondria) was strongly associated with mitochondrial dysfunction. Pre-incubation with the cardiolipin-stabilizing drug elamipretide and raising mitochondrial NAD+ levels both boosted NADH-linked respiration.ConclusionMitochondrial dysfunction is explained by cardiomyocyte architecture disruption and is linked to septal hypertrophy in genotype-negative HCM. Despite severe myocardial remodelling mitochondria were responsive to treatments aimed at restoring respiratory function, eliciting the mitochondria as a drug target to prevent and ameliorate cardiac disease in HCM. Mitochondria-targeting therapy may particularly benefit genotype-negative patients with HCM, given the tight link between mitochondrial impairment and septal thickening in this subpopulation.

This article examines how the human perception of knowledge is structured in the empirical world. It is often argued by scientists that facts in this empirical world can be perceived, which makes us believe that this world is an objective world. However, the human way of making sense of the world is individual and embodied, which causes the creation of an individual world for every human: a body-world. The empirical world is in this case a shared space for multiple bodies that agree on the causality of certain events and objects in that space. Every body-world therefore has its own partial perspective on the knowledge in this shared space, which is formed by the physiology of the body, the cultural background, and the identity of the person. The theater has the power, through the techniques of re-enactment and disruption, to give its audience insight in other situated knowledges from different partial perspectives. It can therefore connect different situated knowledges and create ecological knowledge: the awareness of the connected network of knowledges that is produced in various body-worlds on what is happening in the shared space. Only then can we emancipate knowledge and embrace the various partial perspectives that this shared space of body-worlds has to offer.
