Detection and identification of body fluids are crucial aspects of forensic investigations, aiding in crime scene reconstructions and providing important leads. Although many methods have been developed for these purposes, no method is currently in use in the forensic field that allows rapid, non-contact detection and identification of vaginal fluids directly at the crime scene. The development of such technique is mainly challenged by the complex chemistry of the constituents, which can differ between donors and exhibits changes based on woman’s menstrual cycle. The use of fluorescence spectroscopy has shown promise in this area for other biological fluids. Therefore, the aim of this study was to identify specific fluorescent signatures of vaginal fluid with fluorescence spectroscopy to allow on-site identification. Additionally, the fluorescent properties were monitored over time to gain insight in the temporal changes of the fluorescent spectra of vaginal fluid. The samples were excited at wavelengths ranging from 200 to 600 nm and the induced fluorescence emission was measured from 220 to 700 nm. Excitation and emission maps (EEMs) were constructed for eight donors at seven time points after donation. Four distinctive fluorescence peaks could be identified in the EEMs, indicating the presence of proteins, fluorescent oxidation products (FOX), and an unidentified component as the dominant contributors to the fluorescence. To further asses the fluorescence characteristics of vaginal fluid, the fluorescent signatures of protein and FOX were used to monitor protein and lipid oxidation reactions over time. The results of this study provide insights into the intrinsic fluorescent properties of vaginal fluid over time which could be used for the development of a detection and identification method for vaginal fluids. Furthermore, the observed changes in fluorescence signatures over time could be utilized to establish an accurate ageing model.
Inulin is a soluble dietary fibre, also classified as a prebiotic, extracted from chicory roots. The present study aimed to determine the effect of consumption of native chicory inulin on the stool frequency of middle-aged to older adults (40–75 years old) with uncomfortably but not clinically relevant low stool frequency, specified as two to four days without bowel movements per week. Two randomised, double blind, placebo-controlled crossover trials were conducted using similar protocols in differing populations. Trial A was conducted in Amsterdam, The Netherlands and subsequently Trial B was conducted in Newcastle, United Kingdom. Both trials involved supplementation for 5 weeks with 10 g per day of inulin or placebo, a washout period of 2 weeks, and then crossed over to receive the other treatment. In Trial B, faecal gut microbiota composition was assessed using 16S rRNA gene sequencing. In Trial A, which 10 volunteers completed, the stool frequency was significantly increased to an average 4.9 ± 0.23 (SEM) times per week during inulin periods versus 3.6 ± 0.25 in the periods with placebo (p = 0.01). In contrast, in Trial B which 20 volunteers completed, there was no significant effect of the inulin on stool frequency (7.5 ± 2.1 times per week with inulin, 8.1 ± 3.0 with placebo, p = 0.35). However, many subjects in Trial B had a stool frequency >5 per week also for the placebo period, in breach of the inclusion criteria. Combining the data of 16 low stool frequency subjects from Trials A and B showed a significant effect of inulin to increase stool frequency from 4.1 to 5.0 per week (p = 0.032). Regarding secondary outcomes, stool consistency was significantly softer with inulin treatment compared to placebo periods, it increased 0.29 on the Bristol stool scale (p = 0.008) when data from all subjects of Trials A and B were combined. No other differences in bowel habit parameters due to inulin consumption were significant. None of the differences in specific bacterial abundance, alpha or beta diversity were significant, however the trends were in directions consistent with published studies on other types of inulin. We conclude that 10 g per day of native chicory inulin can increase stool frequency in subjects with low stool frequency.

Understanding the factors that may impact the transfer, persistence, prevalence and recovery of DNA (DNA-TPPR), and the availability of data to assign probabilities to DNA quantities and profile types being obtained given particular scenarios and circumstances, is paramount when performing, and giving guidance on, evaluations of DNA findings given activity level propositions (activity level evaluations). In late 2018 and early 2019, three major reviews were published on aspects of DNA-TPPR, with each advocating the need for further research and other actions to support the conduct of DNA-related activity level evaluations. Here, we look at how challenges are being met, primarily by providing a synopsis of DNA-TPPR-related articles published since the conduct of these reviews and briefly exploring some of the actions taken by industry stakeholders towards addressing identified gaps. Much has been carried out in recent years, and efforts continue, to meet the challenges to continually improve the capacity of forensic experts to provide the guidance sought by the judiciary with respect to the transfer of DNA.
Worldwide, coral reefs are rapidly declining due to increased sea water temperatures and other environmental stresses (Figure 1). To counter the extinction of major coral reef building species on the island of Bonaire, the non-profit organization Reef Renewal Foundation Bonaire is restoring degraded reef sites using corals that are grown in local nurseries. In these nurseries, corals are propagated on artificial trees using fragmentation. After 6-8 months of growth in the nursery, the corals are transplanted to degraded reef sites around the island. Over the years more than 21.000 corals have been outplanted to reef restoration sites in this way. These corals show high survivorship under natural reef conditions but remain under threat by environmental disturbances, such as increased water temperatures, diseases, and competition with macroalgae. A promising intervention to increase reef persistence and resilience is to manipulate the coral-associated microbiome. At present, the composition of the microbiome in nursery-reared and outplanted corals on Bonaire is unknown. The aim of the current project is to identify and isolate naturally occurring beneficial bacteria that may stimulate the resilience of these corals. Our key objectives are: 1) to assess the presence of functionally beneficial bacteria in corals in nursery and restoration sites on Bonaire using metagenomic screening. 2) to design culture strategies to isolate these functionally beneficial bacteria. In the future, a selection of these beneficial bacteria can be applied to the corals to increase their resilience against environmental disturbances.
“Authentieke Vanille uit de kas” richt zich op het ontwikkelen van producten en praktische richtlijnen voor glastelers voor een hogere opbrengst van in de kas geteelde Vanillepeulen uit gezonde planten met meer Vanille en een authentieke geur en smaak. Het hiervoor benodigde onderzoek staat onder leiding van het lectoraat Biodiversiteit en Generade, het Center of Expertise Genomics van Hogeschool Leiden. In het project participeren kennisinstellingen, MKB-bedrijven uit de tuinbouw en bedrijven met expertise in chemie, bodemverbetering en genomics. Het project bestaat uit vier deelonderzoeken: 1. Het bestuivingsonderzoek richt zich op ontwikkeling van een instrument voor optimale overdracht van pollen in Vanillebloemen. 2. Een inventarisatie van het microbioom in Vanilleplanten en -peulen in het wild uit het regenwoud in Costa Rica, plantages op Reunion en Nederlandse kassen zal meer inzicht geven in de correlatie tussen de samenstelling van deze microben en het Vanillegehalte van de peulen en hun lokale geur en smaak. De inventarisatie zal gedaan worden met Next Generation DNA metabarcoding van markers ontwikkeld voor schimmels en bacterieen. 3. Daarnaast richt het onderzoek zich op de fermentatie van de Vanillepeul. Er zal worden onderzocht welke combinatie van microben leidt tot productie van een zo hoog mogelijk gehalte aan precursors van Vanille in groene peulen en een authentieke geur en smaak. Dit wordt gedaan met een combinatie van experimenteel teeltonderzoek en chemische analyses. 4. Momenteel wordt aantasting door schimmels en virussen in de kas met de hand verwijderd. Deze behandeling is kostbaar maar voor biologische teelt de enige optie. Een vierde pijler van het onderzoek zal daarom gericht zijn op de verbetering van het substraat. Met behulp van experimenteel onderzoek zal worden nagegaan welke mix aan microben in het substraat nodig is voor bescherming tegen aantasting door kwaadaardige schimmels.
Lipids, proteins and biological active compounds that are present in insects can serve as nutrient source for poultry production. Because of the potential benefit effects of using insects as feed additives, we must consider the effects of gut microbiome on the insect affects itself, and the expected effect on the microbiome of the broilers that consume these insects. This is specifically important in the situation where live insects are fed to poultry, without prior processing. In this proposal we describe to study whether larvae fed to broilers will affect their microbiome in a positive way for practical applications in poultry industry. Hence, a pilot proof-of-concept study will be carried out as basis for a follow-up proposal for a larger project in the future, that we also like to set-up within this project. In that follow-up proposal, focus will be on the effect of different substrates for insects, on the insect microbiome, to spike insects with specific bacteria and to track their microbiome dynamics over time, and the effect of these insects used as a feed additive on the broiler gut microbiome. This study will provide results on if live Black Soldier Fly larvae (BSFL) can affect the broiler gut microbiome in a positive way, and relevant outcomes will be exploited in a follow-up research proposal in which these effects will be unraveled in detail for adoption by the industry. The project is a collaboration between cooperative insect company RavenFeed and NGN Pro-active both with knowledge on BSFL rearing, Wageningen Bioveterinary Research (WBVR) with knowledge on insect diseases and microbiome analysis, Schothorst Feed Research (SFR) highly experienced in poultry nutrition research and having unique poultry facilities, and Aeres University of Applied Sciences Dronten (AHD) with research facilities for BSFL rearing under experimental conditions.
