BackgroundGait analysis has been used for decades to quantify knee function in patients with knee osteoarthritis; however, it is unknown whether and to what extent inter-laboratory differences affect the comparison of gait data between studies. Therefore, the aim of this study was to perform an inter-laboratory comparison of knee biomechanics and muscle activation patterns during gait of patients with knee osteoarthritis.MethodsKnee biomechanics and muscle activation patterns from patients with knee osteoarthritis were analyzed, previously collected at Dalhousie University (DAL: n = 55) and Amsterdam UMC, VU medical center (VUmc: n = 39), using their in-house protocols. Additionally, one healthy male was measured at both locations. Both direct comparisons and after harmonization of components of the protocols were made. Inter-laboratory comparisons were quantified using statistical parametric mapping analysis and discrete gait parameters.ResultsThe inter-laboratory comparison showed offsets in the sagittal plane angles, moments and frontal plane angles, and phase shifts in the muscle activation patterns. Filter characteristics, initial contact identification and thigh anatomical frame definitions were harmonized between the laboratories. After this first step in protocol harmonization, the offsets in knee angles and sagittal plane moments remained, but the inter-laboratory comparison of the muscle activation patterns improved.ConclusionsInter-laboratory differences obstruct valid comparisons of gait datasets from patients with knee osteoarthritis between gait laboratories. A first step in harmonization of gait analysis protocols improved the inter-laboratory comparison. Further protocol harmonization is recommended to enable valid comparisons between labs, data-sharing and multicenter trials to investigate knee function in patients with knee osteoarthritis.

A large, recently published, inter-laboratory study by the ReAct group has shown that there is considerable variability in DNA recovery that exists between forensic laboratories. The presence of this inter-laboratory variability presents issues when one laboratory wishes to carry out an evaluation and needs to use the data produced by another laboratory. One option proposed by the ReAct group is for laboratories to carry out a calibration exercise so that appropriate adjustments between laboratories can be made. This will address some issues, but leave others unanswered, such as how to make use of the decades of transfer and persistence data that has already been published. In this work we present a method to utilise data produced in other laboratories (whether it provides DNA amounts or a probability of transfer) that takes into account inter-laboratory variability within an evaluation. This will allow evaluations to continue, without calibration data, and ensures that the strength of findings is appropriately represented. In this paper we discuss complicating factors with the various ways in which previous data has been reported, and their limitations in supporting probability assignments when carrying out an evaluation. We show that a combination of producing calibration information for new data (as suggested by the ReAct group) and development of strategies where calibration data is not available will provide the best way forward in the field of evaluations given activities.

from the article: "Cow's milk-derived whey hydrolysates are milk substitutes for cow's milk allergic infants. Safety assessment of these hydrolysates is crucial. Currently, huFcεRIα-RBL-2H3 cells, sensitized with serum IgE from cow's milk allergic patients, are used to assess in vitro residual allergenicity. However, limited availability and high inter-lot variation of sera impede the standardization of safety testing. Recently, we generated an oligoclonal pool of chimeric human (chu)IgE antibodies against bovine β-lactoglobulin (BLG) as an alternative for human serum. These antibodies demonstrated increased sensitivity, specificity and reproducibility. An inter-laboratory ring trial using our new degranulation assay with different whey-based hydrolysates was performed at four independent laboratories to investigate the robustness and reproducibility. RBL-2H3 cells expressing huFcεRIα were sensitized with our oligoclonal pool of anti-BLG chuIgE antibodies. The cells were subsequently incubated with an amino-acid based formula (AAF), two extensively hydrolyzed formulas (eHF) and three partially hydrolyzed formulas (pHF) to assess the degranulation upon challenge. Results demonstrated a very strong inter-laboratory correlation and the intra- and inter-laboratory variations were acceptable. The AAF and both eHFs showed no degranulation, whereas all pHFs demonstrated degranulation. The study showed that this degranulation assay is robust and reproducible within and between laboratories. This new in vitro degranulation assay seems predictive for allergenicity outcome and might therefore be considered as a relevant substitute for animal models."

Elk jaar organiseert de HBO-I stichting, het samenwerkingsverband van hbo ict-opleidingen in Nederland, een conferentie voor haar leden. Tijdens deze conferenties staan strategisch beleid, deskundigheidsbevordering en samenwerking centraal. Sinds 2005 is de internationale oriëntatie als strategisch aspect benadrukt. De blik tien jaar vooruit om de curricula niet op hypes, maar op trends te sturen. In 2005 werd Silicon Valley bezocht en in 2006 ging het HBO-I naar de CeBit in Hannover. Dit jaar zochten we de ict-wijsheid in het verre oosten. Hoe ontwikkelt ict zich in Azië? Dat was de reden voor het HBO-I om een studiereis te maken naar Japan. In Tokyo tref je alle grote ict-bedrijven en drie van de beste Japanse universiteiten. Geen betere periode om Tokyo te bezoeken dan de week van de kersenbloesem. Standard SARS-CoV-2 testing protocols using nasopharyngeal/throat (NP/T) swabs are invasive and require trained medical staff for reliable sampling. In addition, it has been shown that PCR is more sensitive as compared to antigen-based tests. Here we describe the analytical and clinical evaluation of our in-house RNA extraction-free saliva-based molecular assay for the detection of SARS-CoV-2. Analytical sensitivity of the test was equal to the sensitivity obtained in other Dutch diagnostic laboratories that process NP/T swabs. In this study, 955 individuals participated and provided NP/T swabs for routine molecular analysis (with RNA extraction) and saliva for comparison. Our RT-qPCR resulted in a sensitivity of 82,86% and a specificity of 98,94% compared to the gold standard. A false-negative ratio of 1,9% was found. The SARS-CoV-2 detection workflow described here enables easy, economical, and reliable saliva processing, useful for repeated testing of individuals.

Sustainable urban drainage systems (SuDS) such as swales are designed to collect, store and infiltrate a large amount of surface runoff water during heavy rainfall. Stormwater is known to transport pollutants, such as particle-bound heavy metals, which are known to often accumulate in the topsoil. In this study, a portable XRF instrument is used to provide in situ spatial characterization of soil pollutants. The method uses portable XRF measurements of heavy metals along profiles with set intervals (1 meter) to cover the swale with cross-sections, across the inlet, the deepest point and the outlet. Soil samples are collected, and the In-Situ measurements are verified by the results from laboratory analyses. Stormwater is here shown to be the transporting media for the pollutants, so it is of importance to investigate areas most prone to flooding and infiltration. This quick scan method is time and cost-efficient, easy to execute and the results are comparable to any known (inter)national threshold criteria for polluted soils. The results are of great importance for all stakeholders in cities that are involved in climate adaptation and implementing green infrastructure in urban areas. However, too little is still known about the long-term functioning of the soil-based SuDS facilities.

Ergens in 2007, schat men, werd het punt gepasseerd dat 50% van alle wereldbewoners in steden woonde. Dit percentage zal in de 21e eeuw nog sterk toenemen. Zowel het landschap als de stedelijke ruimte staan onder druk, transformeren onder invloed van economische, politieke, sociale of natuurlijke krachten. De huidige groei vindt voornamelijk plaats in ontwikkelingslanden en in de opkomende economieën van China, India en Zuid-Amerika. Een belangrijk deel van de verstedelijking hier is ongepland en ongestructureerd. In Europa doet zich een ander fenomeen voor. Onder invloed van globalisering en de EU beconcurreren de vele steden in het dichte stedelijke netwerk elkaar als vestigingslocatie voor kennisintensieve bedrijvigheid en creatief' talent. Steden en stedelijke regio's transformeren tot kennisregio's en hun dynamiek wordt bepaald door niet-plaatsgebonden, maar grens- en tijdoverschrijdende netwerken. De bijpassende netwerksamenleving is veel minder dan voorheen 'maakbaar' vanuit het oogpunt van overheidshandelen. Individuele burgers, maatschappelijke organisatie en marktpartijen organiseren zichzelf in steeds wisselende bondgenootschappen. Stedenbouw in deze condities betekend vooral het ruimtelijk anticiperen op dynamische maatschappelijke processen. Er is behoefte aan een nieuwe oriëntatie en een instrumentarium om in deze condities succesvol als ruimtelijk ontwerper te opereren. Het lectoraat wil hieraan een bijdrage leveren en zal zich zowel richten op onderzoek naar concepten van stedelijkheid als op toegepast (ontwerpend) onderzoek naar transformatieopgaven in Europese steden.

There has probably never been such an intense debate about the layout of the countryside as the one that is currently raging. There are serious concerns about the landscape, which is being rapidly transformed by urbanization and everything associated with this process, and not only in the Netherlands but also far beyond its borders. Everyone has something to say in this society-wide debate, from local to national governments, from environmental factions to the road-user's lobby, and from those who are professionally involved to concerned private parties. In many cases it is a battle between idealized images and economic models, between agricultural reality and urban park landscapes, between ecological concerns and mobility. This issue of OASE explores the potential significance of architectonic design for transformation processes on the regional scale. Besides considering the instruments that are available to the designer to fulfil this task, the authors also consider how the design can exercise a 'positive' influence on such processes. The various contributions shed light on the potential significance of territory in contemporary design practice and offer critical reflection on the topical discourse that has evolved over recent years.
